Abstract

NADPH is a key factor determining cellular reducing capacity and reductive biosynthesis. However, there has been little study that determines the effects of NADPH on cellular survival and cellular functions. The goal of this current study is to determine the effects of NADPH treatment on survival of cancer cells. Our study using LDH assay showed that treatment of 10 to 1000 micromolar NADPH for 24 hrs or 48 hrs can decrease the survival of C6 glioma cells by 15 to 50% or 5 to 65%, respectively. Trypan blue assays by using a Cell Viability Analyzer have also shown that NADPH treatment can induce significant necrosis of cancer cells. Collectively, our studies have provided first evidence indicating that NADPH can induce necrosis of such cancer cells as C6 glioma cells, suggesting a novel property of NADPH as well as a new means by which cancer cell death can be induced (Supported by the Key Research Grant of Shanghai Municipal Education Committee (to WY), Pujiang Scholar Program Grant (WY), and Shanghai Med‐X Engineering Center Grant of Equipment and Technology of Physical Therapy for Major Diseases (WY)).
