Abstract

The possibility of the use of near infrared (NIR) diffuse reflectance spectroscopy for the estimation of mycelial weight in rice Koji was investigated. NIR reflectance spectra were collected from rice koji samples dried at 100°C for 60 min and ground for 3 min. By multiple linear regression analysis, the results were correlated with the mycelial weight measured by enzymatic hydrolysis, used as the reference method, and a calibration equation was developed. The standard error of prediction was calculated to be 0.31 (mg/g) in the validation of the calibration equation, and the standard deviation of measurements by the reference method was 0.2 (mg/g). These results suggested that the NIR method could be used for the estimation of mycelial weight in rice koji. Modification of the method used to dry koji samples shortened measurement time. By the NIR method, the mycelial weight of rice koji can be measured in 30 min without chemical or biochemical disruption of the rice koji.
