Abstract

Mass regenerated shoots of tissue-cultured shoot primordia from shoot apices in Narcissus tazetta L. var. chinensis ROEM. were cultured on the agar medium containing 0. 2 mg/l NAA and 0.2mg/l BAP at 25°C under continuous lighting condition. At an 8% sugar concentration, shoots formed bulbils and the survival rate was higher. At a 2% sugar concentration, better leaf elongation was observed.Scale segments obtained from shoots that are propagated on medium containing a 3% sugar concentration with 2mg/l NAA and 4 mg/l BAP were cultured before acclimatization on agar medium containing 0.2mg/l NAA and 0.2mg/l BAP. The total fresh weight after culture was greatest at 3% sugar concentration and weight tended to decrease as sugar concentration increased. Total leaf number, shoot number with leaf and length of longest leaf were better at sugar concentration of 3% and 4%. After low temperature treatment, the masses of shoots were planted in pots and acclimatized. Masses cultured at the 8%-12% sugar concentration showed a high survival rate and the total weight of bulb was greater.It was concluded that 18 weeks culture on 8% sugar containing medium was suitable for the growth after acclimatization.
