Abstract

Effects of NAA and BAP on differentiation and growth of shoots in scale culture of Narcissus tazetta L. var. chinensis ROEM. in vitro were investigated.The cultures obtained from floral stem culture and scale culture were treated at 5°C for 47 days, and then cut longitudinally with basal plate. The bulblet segments were cultured on MS medium modified by Ziv et al. (1970) containing various concentrations of NAA and BAP, 3%sucrose and 0.8% agar at 25°C for 71 days.The shoot number was increased in the medium containing 0.22mg/l NAA and 410 mg/ l BAP. The shoot development was better in the medium containing 0.22mg/l NAA and 0.2mg/l BAP. The continuous multiplication system using these two kinds of media is discussed.
