Abstract

Existing software has been expanded to extract full information from UV spectra collected with a photodiode array detector in liquid chromatography. A four-step operator-interactive strategy is developed to establish the elution order of an unknown sample, a situation encountered in lattice design optimization of LC separations. First, a maximum absorbance plot is used to divide the chromatogram in base line separated peak groups. Second, the minimum number of solutes in each group is established from elution profiles at selected wavelengths. Third, representative spectra for each solute are stored at selected elution times. Finally, the spectra from different chromatograms are matched. Examples demonstrate that the composite profiles of severely overlapping solutes can be unraveled by judicious human interpretations. 18 references, 5 figures, 1 table.
