Abstract

The morphological change of a neuroblastoma cell line, Neuro2a, induced by Propentofylline, bifemelane, idebenone and indeloxazine was examined. Neuro2a(a murine neuroblastoma cell line)showed dose-dependent neurite extension. Propentofylline markedly increased the intracellular cyclic AMP level of Neuro2a cells, suppressed DNA synthesis(67% at 1 mM), and induced multipolar(multiple-neurite-bearing)-type neuritogenesis. A similar type of neurite outgrowth was. induced by 8-bromo-cyclic AMP, which also elevated the intracellular cyclic AMP concentration. Bifemelane, idebenone and indeloxazine had also the multipolar-type neuritogenic effects. They, however, failed to induce these morphological changes more than 1 mM, probably owing to the toxic potency of these compounds as compared with propentofylline.
