METHODS Sixteen male Wistar rats were randomly assigned into sham operation and varicocele groups. Left testes were harvested 30 days after the induction of varicocele. Proteomics assaywas performed to determine the expression level of protein of interest. Among the 21 proteins identified expressing differently between sham an varicocele, the levels of 14-3-3∈, Bax, twokey regulators for survival and apoptosis, were detected in germ cellsby western blotting and their location was determined by Immunohistochemistry (IHC). TUNEL method was used to analyze the apoptotic rate of germ cells. In vitro study was also applied to confirm whether silencing 14-3-3∈ can induce apoptosis in GC-1 spg cells. RESULTS Compared with control group, the 14-3-3∈ protein expression was significantly declined while Baxwasincreased in experimental group. The apoptosis index (AI) in experimental groupwas obviously higher than that in sham group. Knockdowning 14-3-3∈ by siRNA greatly induced apoptosis of GC-1 spg cell, with the up-regulation of cleaved caspase-3. CONCLUSIONS Our in vivo study demonstratedthat varicocele caused a significant down-regulation of 14-3-3∈ butup-regulation of Bax, which locally occurred in theapoptoticspermatogenic cell. In vitro study supported that downregulation of 14-3-3∈ contributed to germ cell apoptosis.
