Nevertheless, to our knowledge, there are no studies on the effects of vitamin D deficiency through life (gestational and postnatal periods) on penis morphology. Therefore, this study aimed to evaluate the effects of maternal and postnatal vitamin D deficiency on penis morphology of Wistar rats at 4 months of age. METHODS Wistar females (six weeks of age; ± 270g) were randomized divided into two groups: SC (n = 8, standard chow AIN93G diet - control) or VitD- (n = 9; AIN93G diet without vitamin D) during six weeks pre-pregnancy. After this period, females were mated with males of the same age who received SC diet throughout life. Dams were evaluated for fasting plasma glucose and total weight gain. Offspring were analyzed at 4 month of age for dietary intake, body mass (BM), nasoanal length (NAL), and oral glucose tolerance test. At euthanasia, the penis was dissected for histomorphometrical analyzes. Data were analyzed by unpaired t-test, considering p<0.05. RESULTS Dams did not present any difference in weight gain and fasting glucose in both groups. In the offspring at birth, BM, NAL or glucose did not differ among groups. However, at 4 months of age, the VitD- group had impaired glucose tolerance (+11%; p=0.01), increased adiposity (+40%; p=0.005), but no difference in BM. Regarding offspring penis morphology, both groups showed equivalent Total Area of the Penis and Corpus Cavernosum with Tunica Albuginea (TA). Nevertheless, the Area of TA was lower in VitD- group than SC (-6%; p=0.0003). Additionally, VitD- group showed reduction in the density of smooth muscle (-27%; p<0.0001) and sinusoids (-18%, p=0.008). Inversely, VitD- presented higher volume density of the connective tissue (+23%; p<0.0001) than its counterpart. CONCLUSIONS Vitamin D restriction during peri and postnatal periods impairs metabolic parameters in adulthood. Moreover, it contributed to penile morphological changes in offspring, mainly with lower area of tunica albuginea and density of smooth muscle.
