Better understanding of the molecular mechanisms driving RCC progression may contribute to development of more effective therapeutic and diagnostic intervention strategies. Versican, a large extracellular matrix proteoglycan, is thought to generate a hospitable microenvironment for tumor invasion and metastasis in breast and prostate tissues, as well as many other cancer types. However, information regarding versican in RCC is limited. In this study, we investigated the association of versican expression with clinical parameters in RCC cases. METHODS The expression of versican was analyzed using 3 renal tumor cell lines (Caki-2, ACHN, 786-O), and 84 matched clear cell RCC and normal renal tissues. In addition to performing immunoblotting, and reverse transcription and quantitative PCR assays, we also examined whether down-regulation of versican had an effect on the viability, or migratory and invasion properties of RCC cells. All study protocols were approved by the ethics committee of Shimane University Faculty of Medicine in accordance with the 1975 Declaration of Helsinki. RESULTS The mRNA expression of versican in the RCC cell lines was significantly higher than in the nonmalignant HK-2 cell line and also significantly higher in RCC samples as compared to normal kidney samples (p<0.0001). The higher expression of versican was significantly correlated with high stage disease and systematic metastasis (p<0.05). When the expression level of the versican mRNA transcript was divided into 2 groups based on median values, higher expression was significantly associated with overall survival (OS) after a radical nephrectomy (p=0.0235). After transfection of versican siRNA, cell viability, migration, and invasion ability were significantly inhibited in the RCC cell lines.
