Abstract

Several points are made in the letter by Woodrow and Bustamante [1] regarding our Opinion article on the mechanism of action of artemisinins [2]. The first relates to the relevance of our hypothesis to artemisinin resistance. Whereas there are many possible mechanisms of resistance to artemisinins being explored in several model systems, it is worth remembering that only some of them might relate to the specific target for this class of drug. The PfSERCA-artemisinin hypothesis suggests clearly defined directions for future lines of investigation that stimulate thinking about resistance and tolerance mechanisms in parasites, aiming to include the whole of the calcium homeostatic pathway.
