Abstract

Abstract Isoelectric focusing (IEF) has been used to study humic and humiclike substances. As a general interpretation, these substances are fractionated by IEF depending on their molecular weight. However, the nature of humic-substance fractionation is not certain when an IEF run is applied over them. Even if analysis and extraction methods have changed over the years, the above-mentioned interpretation is still the same. In order to check the actuality of the current interpretation, humic substances were separated to different molecular weight cutoff fractions and isoelectric focusing was applied to them. Results revealed that the dependence of IEF profiles on molecular weight is not a generally valid hypothesis, even if the lowest fraction (1,000–10,000 Da) of molecular weight showed that the smallest amount of material focused on the cathode side in over 50% of the samples studied.
