Abstract
ABSTRACTA new proposed approach for estimation of cosmetic collagen preparations consists on standardization of the gelchromatographic columns in the usual way, subsequent equilibration of the columns with buffer containing 5 molar urea and gel chromatography of the collagen samples in the urea-containing buffer. It is shown that suitable distribution of collagen fragments in a preparation designed for cosmetic purposes could be: 25–30% under 17000Da, 15–25% between 17000 and 140000Da and 50 -60% above 140000Da. With respect to the content, a mean deviation of 5 to 10 residues per 1000 amino acid residues could be acceptable when the contents of a collagen preparation and that of the α1-chain of calf skin collagen type I are compared. It was found that the subjection of the collagen fragment mixture to suitable adsorption methods removes allergenic and pyrogenic substances from it.
