Abstract
In order to obtain oligonucleotide aptamers which can specifically recognize the cytotoxin ricin protein,we designed and synthesized an 87 nucleotides ssDNA pool containing 40 random nucleotides flanked by invariant binding regions for PCR primers,which could be used to screen out a specific aptamer for ricin by SELEX (systematic evolution of ligands by exponential enrichment).After several rounds selection,the 38.5% binding ratio of the ssDNA pool bound to ricin protein can be obtained.The results show that the high efficiency of enrichment has been observed by using affinity resin as separation medium.
