Abstract

The dysregulation of macrophage-mediated interleukin-1β (Il-1β) secretion can lead to chronic inflammatory diseases. Il-1β is a cytokine released as a result of the assembly of the inflammasome: a micrometer-size multiprotein complex that triggers the activation of procaspase-1. Inflammasome assembly is mediated by protein sensors that detect danger signals, and the adaptor protein ASC that tethers sensor and procaspase-1 molecules. The 3D structure of ASC reveals two Death Domains, PYD and a CARD, connected by a flexible linker.
