Abstract

ABSTRACTIn order to elucidate the role of different protein kinase in regulation of cellular responses to interferon alpha and interferon gamma, two mammalian epithelial cell lines—one normal, WISH, and the other cancer, MCF-7, were used. The cell viability was measured by MTT assay after treatment with interferon, protein kinase inhibitor or both. After treatment of WISH cells with interferon alpha or gamma a decrease of the viability of cells was observed. Conversely, the viability of MCF-7 cells treated with the same interferon was increased. Staurosporin manifested a synergistic effect with interferon alpha and gamma on the both cell lines, while PD98059 showed an antagonistic effect. Treatment with Wortmanin increased the effect of interferons in WISH cells and decreased their effects in MCF-7 cells. The cell viability outcomes upon treatment with interferon alpha or gamma in combination with any of the protein kinase inhibitors KN-62, AG490, Gö6983 and Gö6976 were cell type-dependent.
