Abstract

Nonlinear regression analysis based upon the Marquardt algorithm was used to evaluate the suitability of a general enzymic rate equation. The method was illustrated by application to a variety of substrate-modifier mixtures acted upon by α-chymotrypsin. A broad range of kinetic models was readily tested by convenient manipulation of the computer program. The method provides a rapid, quantitative basis for model rejection, thereby permitting efficient coupling of model hypothesis, data collection and evaluation of fit. The methods presented are applicable to any problem in which the experimenter is seeking the best functional relationship between a response and its independent variables.
