Abstract

In a high cell density culture with a recombinant Escherichia coli using a synthetic medium the concentration of glucose as non-limiting substrate was controlled at a certain level in the range between 1.5 and 5 g/1. The glucose concentration was monitored in time intervals of 1 minute using a modified flow-injection-analysis (FIA) device. Due to the high biomass concentration (30...100 g/1) the monitored substrate concentration of the cell suspension differed from off-line estimated values obtained cell free culture supernatant. Using models including indirect measurement of biomass on-line monitored glucose concentration was corrected on-line and was used as the control variable.
