Abstract

Tobacco mosaic virus (TMV) reconstitution in a combination of TMV strain OM (TMV-OM)-RNA and OM-protein or strain B (bean)-protein was carried out at 28°. The reconstitution rate in both reactions was almost the same; however, the mode of reconstitution was different. In homologous reconstitution, two steps occurred and in heterologous assembly three. Infectious TMV particles with mixed protein coats were obtained at various stages of reconstitution. The yield depended upon the amount of protein already coating the RNA molecules. Mixed protein-coated TMV was obtainable. However, in rod formation, interference was also observed.
