Abstract

Hypophysial and preoptic nucleus extracts of yellow and of silver European eels, as granular fractions obtained by ultracentrifugation, were tested for total hormonal activity (arginine-vasotocine + isotocine) by means of the in vitro rat uterus assay according to the method of Holton (1948). The experiments showed that the total hormonal activity is measurable in the hypophysial and preoptic nucleus fractions under certain conditions. An important difference was found in the total hormonal activity of the hypophysial extracts from silver eels and those from yellow eels. In granular fractions it was possible, at least in hypophysial fractions, to show that the total hormonal potency was greater in the neurohypophysial supernatant than in the corresponding granular pellet, if an 0.88 M saccharose solution, buffered with phosphate 10 −2 M at pH 7.4, is used. In the preoptic nucleus granular extracts the values of the activity are more fluctuating.
