Abstract

Objective To investigate the significance of miR-145,and miR-21,the role of miR-10b in breast cancer,providing theoretical and practical foundation for further improving the diagnosis and treatment of breast cancer.Methods Real-time RT-PCR detected 35 cases of breast cancer and adjacent normal breast tissue miR-145,miR-21,miR-10b expression,and analysis of their relationship with clinico-pathological features of breast cancer; the expression was examined by immunohistochemistry in breast cancer ER,PR expression; fluorescencein situ hybridization in detection of HER-2,TOP 2A.Results Expression of miR-145 in breast cancer tissues was lower than in adjacent normal tissue,only 0.78 of normal tissue,but without statistical significance.miR-145 in large lumps,lymph node metastasis,ER-positive,HER-2 positive,TOP 2A-positive patients with low expression (P < 0.05),with age,menstruation,PR expression is independent.Expression of miR-21,miR-10b in breast cancer tissues was significantly higher than that in adjacent normal tissue,which are 8.58,1.70 times the normal tissues (P <0.05).MiR-21 in old age,menopause,lymph node metastasis,HER-2 positive patients with high expression (P <0.05),regardless of tumor size,ER,PR,and the degree of expression of TOP 2A; miR-10b in large lumps,lymph node metastasis,TOP 2A high positive expression (P < 0.05),while with age,menstruation,ER,PR,HER-2 independent.MiR-10b in large lumps,lymph node metastasis,the positive expression TOP 2A high expression (P < 0.05),and with age,unrelated to menstruation,ER,PR,HER-2 expression.Conclusions miR-145,miR-21 and miR-10b in breast cancer development,invasion process may play an important role.
