Abstract

Although in vitro derivation of human primordial germ cell-like cells (PGCLC) is feasible using human embryonic stem cells (hESC), the mechanism of human germ cell specification remained to be investigated. Pluripotency is inhibited by the let-7 microRNA (miRNA) family and promoted by embryonic stem cell-specific cell cycle-regulating (ESCC) miRNA family. We evaluate the role of these two miRNA families in hESC differentiation into somatic lineages versus PGCLC.
