Abstract

Abstract In order to make visible or to photograph structural details of algal chromatophores, an “absorption image” should be produced in the microscope by using a proper light filter. For a number of reasons, this can be done, not by using a red filter whose color is complementary to the green component of the plastid pigments, but by applying a blue filter, complementary to the yellow of both chlorophyll and of carotenoid pigments. The use of Wratten Filter no. 48 is especially recommended for this purpose. Some problems in the use of filters in microscopy are briefly discussed.
