Abstract

In the decade since its introduction, the gene gun has gone from being a scientific novelty to an established tool for introducing DNA and other molecules into plant cells. The unique ability of biolistic technology to deliver macromolecules into intact cells has been exploited in studies ranging from control of gene expression to organelle transformation. Further, biolistics is the critical technology allowing for the genetic transformation of the world’s most important crop species. Still, many of the factors that influence the efficiency of transformation using the gene gun are poorly understood. The purpose of this paper, then, is to review the current research addressing the physical and biological parameters that effect biolistic transformation.
