Abstract

1. 1. Methods currently employed for the solubilization, denaturation, fractionation and identification of cytochrome P-450 from mammalian liver endoplasmic reticulum were compared and evaluated. 2. 2. The omission of a sulfhydryl reducing agent from the pretreatment solution caused changes in the resulting polypeptides resolved upon polyacrylamide gel electrophoresis. 3. 3. Gradient gels had a far greater resolving power than a nongradient gel system when the two types of gels were used to analyze either whole solubilized microsomes or a purified cytochrome preparation. 4. 4. No single selective analytical method was found which could detect all cytochrome P-450-derived polypeptides after gel electrophoresis; however, the isochromic application of samples to the gel was found to facilitate localization of the cytochrome derived polypeptides.
