Abstract

Objective To investigate the effects of Sapylin on proliferation, invasion, autophagy and apoptosis of ovarian cancer cells OVCAR3 and SKOV3. Methods MTT and Transwell were used to detect the effects of Sapylin on proliferation and invasion of OVCAR3 and SKOV3 cells. The effect of Sapylin on cell cycle and apoptosis of OVCAR3 and SKOV3 cells was detected by flow cytometry, and influence of Sapylin on the expression of various tumor suppressor genes in OVCAR3 and SKOV3 cells was detected by Western blot, including cell invasion-related proteins matrix metalloproteinase-2 (MMP-2), cell cycle-related proteins p53 and p27, autophagy-related proteins Atg7 and Lc3II, and apoptosis-related proteins Bax and Bcl-2. Results Sapylin could inhibit the proliferation and invasion of OVCAR3 and SKOV3. And Sapylin could alter the cell cycle of OVCAR3 and SKOV3, by decreasing the proportion of cells in G0/G1 phase and increasing the proportion of cells in S phase, so that promoted cell apoptosis. Sapylin could down-regulate the expression of MMP-2, p53 and Bcl-2, and up-regulate the expression of Atg7, Lc3II, p27 and Bax. Conclusions Sapylin can inhibit the proliferation and invasion of ovarian cancer cells OVCAR3 and SKOV3, change the cell cycle, and promote apoptosis and autophagy.
