Abstract

When α1 adrenergic receptor (α1-AR) binds to the agonist, the G protein-coupled receptor signal is activated, leading to vasoconstriction, a process associated with increased intracellular calcium concentration and activation of the myosin light chain kinase (MLCK) and protein kinase C (PKC)-mediated signaling pathways. In our previous experiments, α1-AR activator phenylephrine(PE) resulted in a significant increase in maximal contraction of the mesenteric artery in elderly rats. However, the expression of α1-AR decreased in these tissues. Therefore, the purpose of this study is to further elucidate the changing mechanism of α1-AR activation leading to vasoconstriction in aging and hypertension.
