Abstract
This chapter discusses the measurement of vasopressin-converting aminopeptidase activity and vasopressin metabolites. Vasopressin (VP) is a nonapeptide with an internal disulfide bond. To study biochemically the significance of VP metabolites, this chapter has devised methods for the characterization and quantitation of VP metabolites as well as the aminopeptidase activity that converts vasopressin to active metabolites. This type of enzyme activity is briefly referred to as VP-converting aminopeptidase activity (VP-AP). The measurement of carboxy-terminal VP metabolites is achieved by virtue of an antiserum that recognizes the carboxy terminus of VP. VP crosses and reacts with the full series of centrally active VP metabolites that are aminopeptidase products. An initial high-pressure liquid chromatography (HPLC) step is used to fractionate tissue extracts into fractions with separated VP metabolites, which are then quantitated by radioimmunoassay. Thus, all carboxyl-terminal VP metabolites are measured individually.
