Publisher Summary This chapter provides an overview of the measurement of the biosynthesis of myo-inositol from glucose 6-phosphate. Myo-inositol, the principal cyclohexanehexol in nature and the precursor of the inositol phospholipids, is synthesized in two stages from glucose 6-phosphate. As in the other fates of glucose 6-phosphate leading to various classes of sugars, the first stage is an isomerization; the product is L-myo-inositol 1-phosphate. The reaction is catalyzed by a specific enzyme, L-myo-inositol-l-phosphate synthase with an absolute requirement for NAD + . The second stage is catalyzed by a phosphatase requiring Mg 2+ . Although the enzyme is distinct from the large classes of nonspecific phosphatases, it cannot differentiate between the enantiomeric forms of myoinositol 1-phosphate. The chapter presents various measurement methods. Myo-inositol 1-phosphate can be measured in several ways, the choice depending on the situation confronting the investigator. The various methods are chemical, enzymatic, and chromatographic.
