Abstract

Measurement of gene expression in individual cells has attracted attention as a tool for understanding the distribution of cellular responses to environmental stimuli. An integrated magnetic force based lab-on-a-chip (Mag-LOC) with a droplet-manipulation system was developed for gene expression analysis in a parallel manner. This system does not need mechanical fluidic devices such as pumps and valves for handling solutions. It can be used as a powerful tool for various biochemical applications by moving and coalescing sample droplets using magnetic beads immersed in mineral oil. The droplet containing magnetic beads and a cell was manipulated with the magnet located underneath the channel, and coalesced with lysis buffer. Using K562 cells as the leukemia model, the cell lyses and reverse transcription (RT)-PCR analysis were successfully performed at a single cell level for beta-2 microglobulin gene.
