Abstract
An enzyme‐catalyzed Diels–Alder reaction in the formation of macrophomic acid can be deduced from the crystal structure of macrophomate synthase. The active site of the Diels–Alderase allows to explain how the single enzyme promotes two carbon–carbon bond formations, two decarboxylations and a dehydration step (see scheme).
