Abstract

Although the monoclonal antibody Ki-67 has obviated the need for counting mitoses as a measure of cellular pro liferation, it is formalin sensitive and works only in fresh frozen sections. The MIB antibodies recognize the same epitope and can be used in both frozen and fixed tissues; however, high temperature antigen retrieval is necessary to unmask the effects of formalin fixation. In this study, we examined the use of low temperature antigen retrieval (LTAR) for unmasking the Ki-67 epitope in rat tissues to restore immunoreactivity of MIB-5. LTAR reduced section detachment without compromising sensitivity. (The J Histotechnol 22:3 19, 1999)
