Abstract

As part of ongoing research into the regeneration physiology of Phalaenopsis, the influence of light quality on embryogenic cell division and endoreduplication levels during the protocorm-like body (PLB) regeneration of Phalaenopsis ‘Spring Dancer’ was investigated. PLBs were induced and proliferated under 5 types of monochrome and combined lighting: red (R60), red30:blue30 (R30B30), red40:blue20 (R40B20), red30:blue15:green15 (R30B15G15), and fluorescent lamp (FL). The highest numbers of PLB were obtained under R30B15G15 irradiation, with explants exposed to R30B15G15 having a higher percentage of cells in the S and G2M phase compared to the other 5 light conditions. The oxidized phenol content in the medium and the activity of stress-related enzymes in the PLBs were also lower in all treatments, except for FL and R30B30. PLBs under R60 irradiation were enlarged and elongated in morphology. Flow cytometry showed the presence of high numbers of endoreduplicated cells (2~32C). A combination of red and green light stimulates embryogenic cell division, with red light potentially being involved in PLB enlargement and cell expansion by the regulation of endoreduplication levels.
