Abstract

We report light activation of DNA methyltransferases (DNMT) through the genetic encoding of a photocaged cysteine. This enables insights into the impact of cancer-related DNMT mutations on in vivo catalysis, enables local and tuneable methylation via DNMT-TAL-effector fusions, and provides insights into methylation-triggered transcriptome dynamics.
