Abstract

ABSTRACT A method was devised for the quantitation of 15α-hydroxyestrone-15-N-acetylglucosaminide (15α-OHE1GNAc), 15α-hydroxyestradiol-15-N-acetylglucosaminide (15α-OHE2GNAc) and their corresponding 3 sulfates, 15α-OHE1SGNAc and 15α-OHE2SGNAc, in late human pregnancy urine. The 4 conjugates were separated and purified by sequential chromatography. Each purified conjugate was hydrolyzed in acid. The steroid was analyzed by fluorescence and the sugar was analyzed by the Morgan-Elson reaction. Internal radioactive standards served to correct for procedural losses, so that accuracy appears to have been achieved. Four subjects were studied. Sulfo-N-acetylglucosaminides predominated. Their ranges in μg/24 hr are: 15 α-OHE1SGNAc, 201–960, and 15α-OHE2SGNAc, 197–464. The N-acetylglucosaminides were determined individually in 2 patients and in combination in the other 2 subjects.
