Abstract

Levan (LVN) is a fructan homopolysaccharide biopolymer which has the unique properties for development of a biosensor platform. Herein, LVN modified disposable electrochemical biosensor was developed for detection of DNA, daunorubicin (DNR) and the biomolecular interaction of DNR with DNA. For this purpose, pencil graphite electrodes (PGE) were used as biosensor platform. The microscopic and electrochemical characterization of LVN-PGEs were done in contrast to the control group and the results were showed that the LVN was successfully modified onto the PGE surface. The optimization of experimental conditions such as LVN concentration and DNA concentration was performed. The detection limit was estimated as 2.74 μg/mL using the equation I = 0.05 x CfsDNA+ 3.54 with the R2 = 0.99 for DNA using LVN-PGEs. DNR-DNA interaction was investigated under the optimum conditions. Robust, sensitive, biocompatible and disposable biosensor surface was achieved by modification of PGE surface using LVN biopolymer for electrochemical monitoring of DNA, anticancer agent, DNR as well as DNA-DNR interaction. The single-use electrochemical DNA biosensor modified with LVN is a model for further studies on the development of novel biopolymers based biosensors.
