Abstract

Liangxue Jiedu Formula is a Chinese medicine formula prepared from medicinal herbs and used in China for the treatment of psoriasis. In order to quickly define the chemical profiles and control the quality of Liangxue Jiedu Formula preparations, a Thermo Q Exactive™ Plus Orbitrap™ mass spectrometer was applied for simultaneous identification of multiple constituents. The sample was filtered through a 0.22μm syringe filter, and 10μl was injected into the LC instrument for LC-MS analysis. In addition, an in vivo mouse model of imiquimod (IMQ)-induced psoriasis-like inflammation was used to investigate the effect of Liangxue Jiedu Formula. Psoriasis severities were assessed daily during the 6-day treatment period using the Psoriasis Area Severity Index (PASI). A total of 110 compounds, including organic acids, phenolic acids, flavonoids, and terpenoids, were identified or tentatively deduced on the base of their retention behaviors, MS and MSn data, or by comparing with reference substances and literature data. To evaluate effects of Liangxue Jiedu Formula on IMQ-induced psoriasis-like symptoms, we assessed redness and scaling and measured skin-fold thicknesses. At the end of the 6-day treatment period, the dorsal skins of IMQ treated mice exhibited the physical phenotype of psoriasis. However, Liangxue Jiedu Formula treatment ameliorated skin scaling, and skin-fold thickness. H&E staining showed Liangxue Jiedu Formula prevented IMQ-induced epidermal thickening, epidermal rete ridge elongation and hyperkeratosis. Thus, an efficient and sensitive method employing ultra-high liquid chromatography coupled with linear trap quadrupole and high resolution mass analyzer-orbitrap was developed for the qualitative analysis of chemical constituents of Liangxue Jiedu Formula. Meanwhile, these findings suggested that Liangxue Jiedu Formula has potential as an alternative, complementary treatment for psoriasis.
