Abstract

Longidorus elongatus and Xiphinema diversicaudatum were labelled by incubation in a dilute aqueous solution of glucose {14C}, both species becoming much more radioactive than Ditylenchus dipsaci. Labelling of microbiological contaminants on the nematode cuticle was minimised by using antibiotics, which did not seem to affect nematode viability. When kept in soil, nematodes lost radioactivity rapidly during the first 18 hr after removal from the glucose {14c}, but thereafter very slowly. Individual nematodes remained readily detectable for at least 12 weeks by direct counting or autoradiographic techniques. Labelled material was found in cuticular and internal parts of L. elongatus and X. diversicaudatum, and 14C was incorporated into both trichloroacetic acid-soluble and acid-insoluble macromolecules. Labelling with 14C seems a valuable technique in studies of the ecology and biochemistry of these nematodes.
