Abstract

UBP34, a soluble 34 kDa protein identified in the moss Physcomitrella patens by photoaffinity labeling with an urea-type cytokinin agonist, belonging to the plant intracellular pathogenesis related (IPR) protein family. This class of proteins is ubiquitous in the plant kingdom but their functions are still unknown. Using a reverse genetic approach we generated knockout mutants at the UBP34 locus by homologous recombination. Knockout plants grown in various conditions do not present any visible phenotypic alteration. However, biochemical and molecular analysis of the knockout transformants reveal the specific photoaffinity labeling of another similar protein.
