Abstract

A simple and reliable method using high performance liquid chromatography had been developed for the determination of kitasamycin (leucomycin A5) residue in swine muscle, swine liver, swine kidney, chicken muscle, chicken liver and egg. The LC separation was performed on a Xterra RP18 column (4.6x150 mm, 5μ) with 0.025 M phosphate buffer solution (pH 2.5)-acetonitrile (7:3, v/v) as mobile phase at the flow rate of 0.8 mL/min, and detected at 232 nm. The target drugwas extracted with 0.2%metaphosphoric acid/methanol(6:4, v/v), and cleaned up with Bond Elut SCX cartridge (500 mg).The recoveries of leucomycin A5 from swinemuscle, swine liver, swine kidney, chicken muscle, chicken liver and egg fortified at 1μg/g was 71.5~79.3%. The detection limit in all tissue samples was 0.05 ppm.
