Abstract
Fluorophores Cy3 and Cy5, when used as labels at the 5′ end of oligonucleotides, enable online kinetic monitoring of template-directed syntheses (see scheme) and self-replicating systems with a high density and accuracy of data. We introduce a method for the direct observation of autocatalytic growth that is based on the simultaneous formation of Cy3- and Cy5-labeled template molecules. The method should be useful for the kinetic screening of catalytic cofactors in microtiter plate readers.
