Abstract

Objective To observe the characteristics of atopie pneumonia in BALB/c mice immunized by keyhole limpet hemocyanin(KLH)and to investigate its mechanisms.Methods Female BALB/c mice were immunized with KLH twice on the 1st day and 7th day.On the 15th day,the mice were sacrificed.Pulmonary tissue sections were made and stained with HE.Splenocyte proliferation was studied using[3H]thymidine incorporation assay.Positive percent of CD4+ and CD8+ T cell in peripheral blood and splenocytes were measured using flow cytometer.mRNA levels of Th1 cytokine IFN-γ and Th2 cytokine IL-4 were evaluated by RT-PCR.The production of IFN-γ and IL-4 in splenocyte supernatant and the specific KLH antibody IgG2a and IgG1 in peripheral blood were detected by ELISA.Results ①KLH can induce chronic pulmonary inflammation in BALB/c mice;②KLH enhanced the proliferation of splenocytes;③KLH activated CD4+ T cells in mice;④KLH induced Th2 dominant response in mice.Conclusions KLH caninduce a chronic atopic pneumonia in BALB/c mice and CD4+ T cell excessively activated and Th2 dominant response may involved in this process.
