Abstract

Objective To study the morphological features,the internal transcribed spacer (ITS) region sequence,and the phylogenetic development of Watsonius watsoni isolated from Macaca fascicularis.Methods The morphological features of W.watsoni were observed with hydrochloric acid carmine staining.The ITS1 and ITS2 were amplified by PCR,and were analyzed by sequencing.The corresponding sequences were downloaded from ITS2 database,and were aligned with the ITS2 sequences in W.watsoni by the ClustalW2,and the phylogenetic trees were constructed by MEGA 4.0.Results The morphological characteristics of W.watsoni from M.fascicularis conformed to the classical taxonomic description of W.watsoni.The obtained ITS1 and ITS2 of W.watsoni by PCR were submitted to GenBank and the accession numbers were KC763806,GU999987,respectively.The length of ITS2 was 295 bp,and the persentage of (G+C) content of ITS2 was 52.20%.The phylogenetic tree based on ITS2 showed that W.watsoni,Homalogaster paloniae,Gastrodiscoides hominis consist of a topology branch,and the bootstrap value is 95%.Conclusion The ITS sequence of W.watsoni was sequenced and the phylogenetic tree based on ITS2 was obtained.
