Abstract
Conditions have been established for the isolation and assay of photosynthetically active chloroplasts and protoplasts from the crucifer Arabidopsis thaliana. The inclusion of 10 mM EGTA or EDTA in the isolation medium is absolutely required for the recovery of active chloroplasts. The average rate of CO 2-dependent O 2 evolution by washed chloroplasts was 97 μmol O 2/mg chl/h. This greatly exceeds the rate of net CO 2 fixation by intact plants under standard atmospheric conditions. Dark storage of Arabidopsis chloroplasts in the presence of free Mg 2+ enhances the stability of photosynthetic capacity. This may distinguish Arabidopsis chloroplasts from those of other species where exposure to free Mg 2+ inhibited subsequent CO 2− dependent O 2 evolution.
