Abstract

The aim of this experiment is to show that the petite-negative yeast Kluyveromyces lactis is unable to form respiratory-deficient mutants with deletions in the mitochondrial genome. However, nuclear mutations in the mitochondrial F1-ATPase can predispose the formation of petite mutants and permit the survival of cells lacking mitochondrial DNA (ϱ°). The mutant F1-AT Pase has therefore gained a novel function that suppresses the lethal phenotype induced by the loss of mtDNA.
