Abstract

Leaf mesophyll protoplasts from sterile shoot cultures of Solanum nigrum were isolated by means of enzymic degradation of the cell walls at a yield of 5·10 5 to 10 6/g fresh weight. No viable protoplasts could be obtained from greenhouse plants. Protoplast division and cell cluster formation occurred in modified KM and in V-KM media. Subsequent culture on various solidified media revealed callus proliferation, organogenesis, and regeneration of whole plants.
