Abstract

The Cibinong Science Center (CSC), Indonesian Institute of Sciences (LIPI), Cibinong is overgrown with various types of trees, which could be colonized by a variety of wood fungi. The aim of this study is to isolate and identify the wood fungi and its mycoparasites in the CSC area. Three wood macrofungi were collected from CSC, namely CSC 20, CSC 21 and CSC 22. Isolation was done using the selective media Rose Bengal Chloramphenicol Agar (RBCA). ITS region of rDNA was amplified using ITS5 and ITS4 primers. Molecular identification was done using Sanger sequencing method. Sequence homology search was done using BLAST program. Sequence alignment was carried out using ClustalW from BioEdit. The genetic relationship was inferred using ClustalX and Neighbor Joining (NJ) method. Genomic DNA of isolates CSC 20a, CSC 20b, CSC 22b and CSC 22c were successfully isolated and amplified. The consensus sequence length of ITS rDNA from four isolates was about 706–773 bp. The results of BLAST sequences homology search showed 94–97 % similarities to their closely related spesies. Sample CSC 20 was mycoparasitized by two isolates; Fusarium sp. (isolate CSC 20a) and Trichoderma harzianum (isolate CSC 20b). Sample CSC 22 was identified as Xylaria feejeensis (isolate CSC 22b), and mycoparasitized by Trichoderma sp. (isolate CSC 22c). Taxonomic position of four fungi isolates were separated to three different groups with 100 % bootstrap value. Isolates CSC 20b and CSC 22c were clustered together in a monophyletic group.
