Abstract

This study was done to identified bacteria of genus Salmonella and Shigella from sumatran orangutan (Pongo abelii) feces. Nine semi wild orangutan fecal samples were obtained in Orangutan Reintroduction Center, Cagar Alam Pinus Jantho, Aceh Besar. The samples were processed using Carter methode. The selective media used were Salmonella Shigella Agar (SSA), Eosin Methylen Blue (EMB), and MacConkey Agar. The separated colonies which grawn in SSA media were observed for their morphology, and followed by Gram staining methode then continued with biochemical test such as Indol, Methyl Red (MR), Voges Proskauer (VP), Sulfid Indol Motility (SIM), Simmons Citrate (SC), and sugar test such as glucose, sucrose, lactose, and manitol. From nine samples, one orangutan was infected with Salmonella and another one was infected with Shigella. All of the samples were identified with Escherichia. In Conclusion, orangutan lived at Reintroduction Center, Jantho infected with Salmonella and Shigella bacteria.Key words: feces, orangutan, Salmonella, Shigella

MATERI DAN METODE

Sampel yang digunakan dalam penelitian ini adalah 9 sampel feses dari 9 ekor orangutan sumatera (Pongo abelii) yang diambil di Pusat Pelepasliaran Orangutan Cagar Alam Pinus Jantho. Prosedur Penelitian Sampel feses diambil dengan menggunakan spatula dan dimasukkan ke dalam botol steril kemudian disimpan di dalam lemari es. Identifikasi dan Isolasi Salmonella dan Shigella Feses dimasukkan kira-kira sebanyak 1 gram ke dalam Nutrient Broth dan diinkubasikan selama 24 jam pada suhu 37° C kemudian ditanam pada media Salmonella Shigella Agar (SSA) dengan teknik goresan T. Koloni terpisah pada media SSA diamati morfologinya (bentuk, diameter, elevasi, tepian, warna, dan konsistensi) dan dilanjutkan dengan pewarnaan Gram. Koloni yang telah diamati secara mikroskopis ditanam pada nutrient agar untuk ditanam pada media eosin methylen, MacConkey agar, uji biokimia indol, methyl red, voges proskauer, sulfid indol motility, simmons citrate (IMVIC), triple sugar iron agar (TSIA), dan uji gula-gula yaitu glukosa, sukrosa, laktosa, dan manitol.
