Abstract

The phenolic fraction obtained from enzymically hydrolysed late-pregnancy urine was subjected to partition chromatography on a silicagel column. The fractions obtained were chromatographed on paper in various systems. By means of the Folin-Ciocalteu reagent, spots corresponding with the mobilities to those of 15β-hydroxyoestrone and 15β-hydroxyoestradiol-17β were detected. The fractions believed to contain 15β-hydroxyoestrone and 15β-hydroxyoestradiol-17β were subjected to preparative-scale chromatography on formamide-impregnated paper, using different solvents as mobile phases. The crystalline fractions thus obtained were further purified on Celite columns and subsequently identified as 1 5β-hydroxyoestrone and 15β-hydroxyoestradiol-17β by melting points, mixed melting points, ultraviolet and infrared absorption spectra, formation of derivatives and various microchemical reactions.
