Abstract
A method has been developed, utilizing mutanolysin and proteinase K, for the rapid lysis of strains of the rumen cellulolytic bacteriumRuminococcus. This has enabled bacterial chromosomal and plasmid DNA to be isolated. A small cryptic plasmid has been identified inRuminococcus flavefaciens strain 186. It is 5.2 kb long, contains a singleBamHI site, and two sites forBglII,EcoRI, andHindIII. This plasmid has potential in the development of genetic vectors for rumen bacteria.
