Abstract

Among the Bacillus subtilis strains resistant to folic acid antagonists, afo mutants are phenotypically thymine independent and their dihydrofolate reductase, though expressed at normal levels, is still sensitive to these inhibitors. afo mutants isolated in our laboratory are all thymine independent and methotrexate resistant over a wide range of concentrations but, like wild-type cells grown in the presence of thiamine, at high doses they return to show methotrexate sensitivity. They are all sensitive to trimethoprim, resistant to oxythiamine and show hypersensitivity to pyrithiamine. Though the phenotypic behavior of these afo mutants is very similar, genetic analysis suggests that they may belong to, at least, two different complementation groups. One of these mutations, for which we propose the name afoA, was mapped.
